1.用含10%小牛和的RPMI-1640培養(yǎng)液將K562細(xì)胞配成1&time105/ml濃度。用同樣培養(yǎng)液將MC配成1&time106/ml濃度。
2.取96孔圓底，每孔加0.1ml K562細(xì)胞懸液;每孔加適量MC使效靶比為1：1～5：1，每種處理3個(gè)復(fù)孔;3個(gè)對(duì)照孔只加效應(yīng)細(xì)胞;8個(gè)對(duì)照孔只加靶細(xì)胞;4個(gè)對(duì)照孔只加細(xì)胞培養(yǎng)液。每孔加20l Alamar Blue;每孔總量為0.2ml。
3.在37℃ 5% CO2的飽和水汽二氧化碳培養(yǎng)箱中培養(yǎng)24小時(shí)。
4.直接在熒光檢測(cè)儀上檢測(cè)熒光強(qiáng)度，激發(fā)波長(zhǎng)530nm，發(fā)射波長(zhǎng)590nm。各孔熒光強(qiáng)度值應(yīng)減去培養(yǎng)液對(duì)照熒光強(qiáng)度的平均值，各復(fù)孔熒光強(qiáng)度的平均值記為AF。按下式計(jì)算特異性殺傷百分比：
